Gamma-Glutamyl Transpeptidases: Structure and Function

ثبت نشده
چکیده

Gamma-glutamyl transpeptidases (c-GTs) belong to the N-terminal nucleophile hydrolase superfamily, enzymes that cleave the c-glutamyl amide bond of glutathione to give cysteinylglycine. The released c-glutamyl group can be transferred to water (hydrolysis) or to amino acids or short peptides (transpeptidation). c-GT plays a key role in the gamma-glutamyl cycle by regulating the cellular levels of the antioxidant molecule glutathione, hence it is a critical enzyme in maintaining cellular redox homeostasis. c-GT is upregulated during inflammation and in several human tumors, and it is involved in many physiological disorders related to oxidative stress, such as Parkinson’s disease and diabetes. Furthermore, this enzyme is used as a marker of liver disease and cancer. This book covers the current knowledge about the structure–function relationship of c-GTs and gives information about c-GT applications in different fields ranging from clinical biochemistry to biotechnology and biomedicine.

Structural basis for the inhibition of Mycobacterium tuberculosis l,d-transpeptidase by meropenem, a drug effective against extensively drug-resistant strains

Difficulty in the treatment of tuberculosis and growing drug resistance in Mycobacterium tuberculosis (Mtb) are a global health issue. Carbapenems inactivate L,D-transpeptidases; meropenem, when administered with clavulanate, showed in vivo activity against extensively drug-resistant Mtb strains. LdtMt2 (Rv2518c), one of two functional L,D-transpeptidases in Mtb, is predominantly expressed over...

Cytosolic action of phytochelatin synthase.

Glutathionylation of compounds is an important reaction in the detoxification of electrophilic xenobiotics and in the biosynthesis of endogenous molecules. The glutathione conjugates (GS conjugates) are further processed by peptidic cleavage reactions. In animals and plants, gamma-glutamyl transpeptidases initiate the turnover by removal of the glutamate residue from the conjugate. Plants have ...

Garlic γ-glutamyl transpeptidases that catalyze deglutamylation of biosynthetic intermediate of alliin

S-Alk(en)yl-L-cysteine sulfoxides are pharmaceutically important secondary metabolites produced by plants that belong to the genus Allium. Biosynthesis of S-alk(en)yl-L-cysteine sulfoxides is initiated by S-alk(en)ylation of glutathione, which is followed by the removal of glycyl and γ-glutamyl groups and S-oxygenation. However, most of the enzymes involved in the biosynthesis of S-alk(en)yl-L-...
